Scientific theme and general objectives

The TheREx team (Experimental Recombinant Therapy) belongs to the Medical and Pharmaceutical Schools of the Université Grenoble Alpes. Its objective is a research from patient’s bed to bench and back. Therefore, TheREx’s research project goes from preclinic to clinic, around the use of Synthetic Biology and its development for patients suffering from oncology and haematology diseases or rare enzymopathy.

Research topics

Research topics are based upon
1) rational design of therapeutic proteins and intracellular transfer vectors, and
2) the bases of interactions between human and his bacterial environment. Those two topics are mutually interacting and backed by a preformuling activity that allows the transformation of an active substance – chemical of biological – in a pharmaceutical substance stable and biodisponible, and an early phase clinical trial platform in onco-haematology.

Rational design of therapeutic proteins and intracellular transfer vectors

We developed four approaches to allow intracellular protein transfer: MD11 domain of ZEBRA protein, proteo-liposomes, the dodecahedron from adenovirus, and type III secretion system of P. aeruginosa. In addition, we designed a strategy to compare efficacies of these systems that we apply to production of induced pluripotent stem cells (iPS).
From this know how and our experience of in vitro protein expression systems or secretion by eukaryote cells, we explore rational design of biologics in the field of rare diseases and anti-tumour therapy. Our candidates biologics are directly tested in vivo in mice with the help of the High R-Technology Animal Facility of Institute Jean Roget.

These developments span from basic research, at the molecular and cellular levels, in order to understand the mechanisms of actions of our targets, to therapeutic research in domains from vaccine production (anti-infectious or anti-tumour) to innovative therapies (cancer and orphan diseases, especially haemophilia and Chronic Granulomatous Disease), and regenerative medicine with the production of iPS from cord blood CD-34+ cells. This last project is in collaboration with the French Blood Bank Service (EFS) and the team of Prof. Luc Douay (Saint Antoine) and aims at the production of therapeutic grade red blood cells in bioreactor.

Two start-ups are derived of our research, Synthelis in 2011 and APCure in 2012, and employ currently 17 persons.

Base of interactions between human and his bacterial environment

This approach is conceived lato sensu and includes haematopoiesis and its disorders (myeloproliferative and myelodysplasic disorders), the molecular pathology of neutrophil polymorphonuclears (Chronic Granulomatous Disease), interkingdom communication between P. aeruginosa and his host, and determination of new therapeutic targets in C. albicans.

Over this period of time the team won €1,523,054 of public funding and €988,345 of private funding totalling €2,511,399.
Over the same period, the team has published 160 articles in peer-reviewed journals, filled 5 patents and recruited 6 faculty researchers (4 professors and 2 assistant professors) and a researcher (CR EFS).

L’objectif de l’unité est de fournir une expertise dans le domaine de l’analyse morphologique à travers l’examen de coupes histologiques mais également d’immunomarquages pour la détection d’antigènes spécifiques.
Une approche quantitative peut également être réalisée en fonction du type d’immunomarquage.
