L’équipe HumProTher, dirigée par le Pr Jean-Luc Lenormand, a été créée en 2005 grâce à un projet qui a été financé par le programme Européen Marie Curie Excellence Grant (FP7). L’équipe HumProTher s’intéresse au développement de nouvelles technologies dédiées à la production et à la vectorisation de protéines médicaments à visées thérapeutiques ou vaccinales. Dans ce cadre, les activités de l’équipe HumProTher sont orientées sur le développement de nouveaux vecteurs peptidiques ou lipidiques pour la transduction de molécules à visée thérapeutique.
Vecteurs lipidiques : Une des approches thérapeutiques originales développée au sein du laboratoire repose sur l’utilisation de liposomes comme vecteurs pour le transport et le transfert intracellulaire de protéines membranaires à visée thérapeutique. Cette approche innovante a pu être réalisée grâce au développement d’un nouveau système de synthèse des protéines membranaires basé sur un lysat bactérien d’Escherichia coli (Brevet # A61K9/127; C07K14/00, US2010189774 (A1). Cette stratégie thérapeutique unique basée sur le développement et l’optimisation d’un système de synthèse des protéines membranaires sous forme native et fonctionnelle a permis la production d’un biomédicament pour le traitement des glioblastomes et d’un vaccin recombinant pour lutter contre les infections à Pseudomonas aeruginosa. Ce procédé de synthèse des protéines membranaires a été aussi valorisé par la création d’une société de services dénommée Synthelis (www.synthelis.fr ) et récompensé par les concours nationaux d’aide à la création d’entreprise dans la catégorie« Emergence » (2009) et « Création-Développement » (2010).
Vecteurs peptidiques : Une approche de vectorisation développée au laboratoire, repose sur l’utilisation d’un peptide de transduction (Cell-Penetrating Peptide) issu de la protéine ZEBRA du virus Epstein Barr. Ce peptide que nous avons isolé et caractérisé est capable de transporter des macromolécules et de permettre leur transfert intracellulaire grâce à un passage direct dans la membrane plasmique des organismes vivants ciblés (champignons, cellules de mammifères). Nous avons déjà réalisé la preuve de concept de l’utilisation de ce peptide sur différentes protéines à visée thérapeutique dont l’activité anti-tumorale était préservée. Ce peptide de transduction permet l’internalisation de ces molécules biologiques dans un grand nombre de lignées de cellules tumorales de diverses origines (Brevet #US201113643598 20110407, US2013116201 (A1). Cette stratégie de transduction est unique par rapport aux autres CPP décrits dans la littérature, car elle s’affranchit du processus d’endocytose qui peut s’avérer délétère pour la molécule biologique internalisée. Le peptide de pénétration cellulaire issu de la protéine ZEBRA représente donc un outil puissant pour le transfert intracellulaire de molécules thérapeutiques. Ce peptide est utilisé aussi dans le cadre de nombreuses collaborations pour le transport de protéines :
- permettant la mise en place d’iPS,
- pour de l’enzymothérapie,
- pour de la dédifférenciation de cellules myogéniques,
- pour de la thérapie anti-tumorale
- pour la conception de vaccins recombinants
- pour le traitement de maladies infectieuses
